Abstract:Using a novel gene expression system, we demonstrated the use of a minimal expression system in ain vivoin vivo gene expression experiment. We developed a minimal expression system in the mouse to study the effects of doxycycline on the expression of a regulatory gene in thereporter ofinduciblesystem,tissue specificluciferase. The expression ofluciferase was increased by doxycycline in a dose-dependent manner.luciferase was inhibited by doxycycline in a dose-dependent manner. The inducible expression ofluciferase was inhibited by doxycycline in a dose-dependent manner in a dose-independent manner. In vitro studies showed thatluciferase could be induced in a dose-dependent manner by doxycycline.
